Le Dr Pelletier et Alexandra Katigbak, \u00e9tudiante dipl\u00f4m\u00e9e, ont r\u00e9cemment fait le point sur les progr\u00e8s de leurs travaux.<\/h3>\n<h3><strong style=\"color: #333333; font-family: 'Droid Sans', 'Helvetica Neue', Helvetica, Arial, sans-serif; font-size: 15px; line-height: 24px;\">Titre<em>&nbsp;:<\/em><\/strong><em style=\"color: #333333; font-family: 'Droid Sans', 'Helvetica Neue', Helvetica, Arial, sans-serif; font-size: 15px; line-height: 24px;\">Validation des g\u00e8nes suppresseurs de tumeurs associ\u00e9s au lymphome de Burkitt<\/em><\/h3>\n<p>Le lymphome de Burkitt (LB) est un type de lymphome non hodgkinien. Il se caract\u00e9rise par la mutation d\u2019un g\u00e8ne appel\u00e9 <em>myc<\/em>, qui d\u00e9clenche la croissance incontr\u00f4lable des cellules du LB. Mais on en sait encore trop peu sur les causes de la maladie. Des \u00e9tudes g\u00e9n\u00e9tiques sur les tumeurs du LB ont mis en \u00e9vidence un certain nombre de g\u00e8nes qui ont subi une mutation dans ces tumeurs. L\u2019objectif de notre recherche consiste \u00e0 \u00e9tablir si ces g\u00e8nes mutants jouent un r\u00f4le dans le d\u00e9veloppement du LB.<\/p>\n<p>Afin de d\u00e9finir le r\u00f4le des g\u00e8nes mutants identifi\u00e9s, nous avons utilis\u00e9 un mod\u00e8le murin du lymphome de Burkitt pour examiner chaque g\u00e8ne. Normalement, ces souris d\u00e9veloppent un lymphome \u00e0 cause d\u2019une mutation du g\u00e8ne <em>myc<\/em>. Il est possible de manipuler les cellules souches du sang pour y inclure l\u2019un des g\u00e8nes mutants, et de les transplanter dans les souris afin de reconstituer leur syst\u00e8me immunitaire. Si la mutation g\u00e9nique contribue \u00e0 la formation de tumeurs, les lymphomes \u00e0 cellules B se d\u00e9veloppent plus rapidement que d\u2019habitude. Si la mutation n\u2019a aucun effet sur la croissance tumorale, les lymphomes \u00e0 cellules B se d\u00e9velopperont \u00e0 un rythme normal.<\/p>\n<p>Pour introduire avec pr\u00e9cision les mutations g\u00e9n\u00e9tiques d\u00e9sir\u00e9es, nous avons eu recours \u00e0 un outil sp\u00e9cialis\u00e9 du g\u00e9nie g\u00e9n\u00e9tique. Au total, 91 g\u00e8nes identifi\u00e9s par le biais d\u2019exp\u00e9riences sur le d\u00e9pistage g\u00e9n\u00e9tique du LB ont \u00e9t\u00e9 implant\u00e9s dans des cellules du lymphome de Burkitt. Ces cellules ont alors \u00e9t\u00e9 transplant\u00e9es dans des souris, et l\u2019on a surveill\u00e9 le temps \u00e9coul\u00e9 jusqu\u2019\u00e0 l\u2019apparition du lymphome. Nous avons r\u00e9alis\u00e9 notre premi\u00e8re s\u00e9rie d\u2019exp\u00e9riences et identifi\u00e9 des mutations g\u00e9n\u00e9tiques qui acc\u00e9l\u00e8rent le d\u00e9veloppement des tumeurs. Les travaux en cours visent \u00e0 confirmer nos r\u00e9sultats, puis \u00e0 caract\u00e9riser la fonction de ces g\u00e8nes. Nous r\u00e9aliserons par la suite d\u2019autres exp\u00e9riences pour savoir comment ces g\u00e8nes contribuent \u00e0 la croissance des tumeurs.
